However, only two allergens have been identified from mango so far. Here the protein crude extracts of mangos were resolved by using 1-D sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) followed by western blot. A predominant reactive band with a molecular mass of 30 kDa was analyzed by MS/MS mass spectrometry. The band contained protein fragments which exhibited significant homology to mango chitinase (GenBank Accession: ACD69683.1). The cDNA of the chitinase was synthesized and cloned into a pET-28a plasmid and expressed as a recombinant His-tagged protein. The reactivity of the recombinant protein toward mango-allergic patients was assayed by using western blot. The linear IgE epitopes of the chitinase were then analyzed by spanning peptides. The mango chitinase reacted with IgE of 9 out of 13 sera (69%) of pediatric patients with mango allergy. Only one peptide (8-28) which contained the chitin-binding domain possessed IgE reactivity.
